The Evolving Role of Special Stains in the Histopathological Diagnosis of Connective Tissue Tumors: A Comprehensive Review

Special histochemical stains remain essential in the diagnosis of connective tissue tumors, where morphological overlap complicates differentiation. This review examines their role in highlighting key matrix components—collagen, reticulin, elastic fibers, and mucins—to aid in distinguishing tumor subtypes. Masson’s trichrome and Van Gieson stains identify collagen-rich tumors, reticulin stains outline vascular and architectural patterns, Alcian Blue detects myxoid matrix, and elastic stains assess vascular integrity. These stains provide rapid, accessible, and cost-effective diagnostic support, guiding the selection of further ancillary tests such as immunohistochemistry and molecular studies. In both routine and resource-limited settings, they enhance diagnostic accuracy and workflow efficiency, ensuring precise tumor classification and optimized patient management. Their continued integration into pathology practice underscores their lasting value in the era of advanced diagnostics.
